چکیده

This work was presented in part at the 53rd Annual Meeting of The American Thyroid Association, Cleveland, Ohio, 7-10 September 1977. Dr. Larsen is an Investigator of the Howard Hughes Medical Institute. Received for publication 29 August 1977 and in revised form 11 January 1978. well with both the change in TSH and the nuclear occupancy, suggesting a linear relationship between the integrated nuclear occupancy by T3 and TSH release rate. The contribution of intrapituitary T4 to T3 conversion to nuclear T, was accompanied by an appropriate decrease in TSH, supporting the biological relevance of nuclear T3. Pretreatment of the animals with 6-n-propylthiouracil before T4 injection decreased neither the nuclear T3 derived from intrapituitary T4 nor the subsequent decrease in TSH. These results indicate that intracellular monodeiodination of T4 contributes substantially to the nuclear T3 in the pituitary of the hypothyroid rat, and suggest a linear inverse relationship between nuclear receptor occupancy by T3 in the pituitary and TSH release rate. The data further indicate that T4 to T3 monodeiodination is considerably more important as a source of nuclear T3 in the pituitary than in the liver and kidney. This provides a mechanism whereby the TSH secretion could respond promptly to a decrease in thyroid secretion (predominantly T4) before a decrease in plasma T3 would be expected to lead to significant metabolic hypothyroidism.

Partial purification of the triiodothyronine receptor from rat liver nuclei. Differences in the chromatographic mobility of occupied and unoccupied sites.

A 60to 125-fold purification of nuclear extracts from rat liver triiodothyronine receptor was achieved by dialysis of 0.4 M NaCl nuclear extracts followed by chromatography on DEAE-Sephadex. Extract was labeled either by injecting tracer [““Iltriiodothyronine into the animal or by direct addition of the tracer in. vitro prior to chromatography. Elution of the column with a linear NaCl gradient ...

Thyroid hormone action in cell culture: domonstration of nuclear receptors in intact cells and isolated nuclei.

Triiodothyronine and thyroxine induce a 3-fold increase in the rate of growth of GH(1) cells in culture. To study further the action of these hormones, we examined the binding of [(125)I]triiodothyronine and purified [(125)I]thyroxine to cellular fractions after incubation with intact cells in serum-free medium. High-affinity, low-capacity binding sites for the hormones were demonstrated in nuc...

Putative L-triiodothyronine receptors in the liver nuclei of mature tropical toad, Bufo melanostictus.

Thyroid hormones exert a major role in growth and differentiation of almost all types of tissues in animals, particularly in amphibian metamorphosis, through its specific nuclear receptor activation followed by gene expression. However, its function in mature tropical amphibians is less studied. The present study revealed the existence of a single class of specific nuclear receptor(s) in the li...

Thyroid Hormone Receptors BINDIXG CHARACTERISTICS AND LACK OF HORRlOn-AL DEPEXDENCY FOR NUCLEAR

Thyroid hormones have diverse effects on growth and metabolism. Specific “receptor” proteins which bind triiodothyronine and other biologically active analogs and which may be involved in thyroid hormone action have been recently found in nuclei of responsive tissues. This report presents studies of these receptors in rat liver nuclei. Confirming previous reports, a Scatchard analysis of the bi...

Thermodynamics of triiodothyronine nuclear receptor interaction. Role of hydrophobicity in triiodothyronine binding to its receptor.

We have investigated the thermodynamic properties of triiodothyronine (T3)-solubilized hepatic nuclear receptor interaction. These studies revealed that the equilibrium constant was markedly dependent on temperature and van't Hoff plots revealed a significant deviation from linearity. Both enthalpy and entropy changes decreased with increasing temperature such that at 37 degrees C both were neg...
